It's more of a cabling convenience thing. I'd go M.2 over traditional SATA any day. Less cables, less fuckery.

The second slot shares bandwidth for the PCIe 3.0 4x slot so if you use it you sacrifice the slot. The first slot is connected to the CPU and second the SB. So yeah, 1x4.0 and 1x 3.0 or 2x 3.0.

That board only looks like it supports one M.2 @ PCIe 4.0.

And have to note not to underestimate Infinite to boot because no one is talking about it.
